標題何謂組織工程(Tissue Engineering)

以下是Arnold I. Caplan的文章中的一段對話(錄自TISSUE ENGINEERING 6:1,2000�
Graduate student:"Give me a great tissue engineering thesis project
that will win you a Nobel Prize and insure that I get a great job when
I leave your laboratory"
Mentor 1 (biologist):"Go study published papers in the library on exactly
how embryos fabricate tissues, write down all of the key events, and use these
as rules of the road to tissue engineer the regeneration of organs or complex
tissues in the adult organism."
Mentor 2 (material scientist):"Go study published papers in the library
on material properties of adult tissues, write down all the key parameters
and characterize srecifi cations, and use these as rules of the road for
designing tissue substitutes whose material properties exactly match those
of the adult tissue."
Mentor 3 (engineer):"Go study published papers in the library on
mechanical properties of tissues, and use these properties as rules of the
road to design a matematical (finite element) model to predict how the
tissue will change with age and change in various modalities."
Mentor 5 (molecular biologist):"Go study published papers in the library
on all of the sequencing information that you can obtain from the Human
Genome Project and elsewhere on the promoter control elements that are
involved in forming and/or repairing different tissues, and use these to
design vectors to transform primitive cells into cells which will fabricate
the tissues we're interested in."
A voice from above:"Combine all of the above and call it tissue
engineering." The abjective of the above is to stress that tissue engineering
is viewed in many different ways by experimentalists depending on their
training/ background and experience. Because tissue repair and regeneration is
a complex, multistep, multicomponent, and highly coordinated process in a very
complex surgical site, all of the various factors, components, parameters, and
considerations must be managed. It took Mother Nature millions of years to
from multicellular organisms with specialized cells, tissues, and rocesses,
and it took Her a long time to select for successful repair motifs in adults.
The mimicking of these fabrication processes must coordinate a large number
of variables; it is hoped that we experimentalists will not take as long to
learn the "tricks" of tissue engineering in order to improve upon Mother
Nature's rules of the road for tissue repair/regeneration.
