Xenical�: Significant weight loss benefits proven in biggest study
ever 

Xenical, Roche's new anti-obesity treatment, promotes significant weight-loss, lessens weight-regain and
improves obesity-related risk factors. These findings of a two-year US clinical study are published in the
Journal of the American Medical Association JAMA. 

In this largest placebo-controlled pharmaco-intervention study in obese people lasting for 2 years, 892
patients were randomly assigned to double-blind treatment. During the first year Xenical-treated patients lost
significantly more weight (8.8%) than placebo-treated patients (5.8%). Patients treated with Xenical 120 mg
three times a day regained significantly less weight during year 2 (35.2% regain) than those receiving placebo
(63.4% regain).

Treatment with Xenical was associated with significant improvements in fasting lipid and insulin levels. The
significant and sustained lowering of insulin levels is clinically important as earlier studies link fasting serum
insulin levels with ischemic heart disease risk, insulin resistance and obesity-related hypertension. These
findings are consistent with those of a European study published recently in The Lancet. In addition to
promoting weight loss, Xenical was shown to improve total and LDL cholesterol beyond what is expected
from weight loss alone in this European study. 

Xenical is the first of a new class of non-systemically acting anti-obesity drugs called lipase inhibitors which
act in the gastrointestinal tract to block the absorption of fat. Xenical does not achieve its effect through
brain chemistry. Final approval has already been achieved in the European Community, in several countries
in Latin America and the Far East and is already marketed in 13 markets with more than 700'000 patients
treated. In the United States, Roche has submitted this week follow-up data from ongoing clinical studies to
the FDA. These data were requested when the FDA granted approvability status to Xenical in May 1998.
